Os objetivos desta revisão foram apresentar o estado da arte das tecnologias utilizadas para desenvolver cultivares em espécies alógamas, excluindo as espécies perenes e as propagadas assexuadamente. Foi revista a estrutura genética destas espécies e suas relações com o desenvolvimento de cultivares, as tecnologias utilizadas para desenvolver híbridos e variedades de polinização aberta melhoradas e seus F1´s, e o impacto da biotecnologia e dos modelos mistos no desenvolvimento de cultivares. Um sumário dos resultados das pesquisas desenvolvidas no projeto de melhoramento de milho do Departamento de Genética da Escola Superior de Agricultura "Luiz de Queiróz" será apresentado.

Objectives of this review were to present the state of the art of the technologies used to develop cultivars in allogamous species, excluding the perennial and asexually propagated species. It was reviewed the genetic structure of these species and its relationship with the development of cultivars, the technologies used to develop hybrids and improved open-pollinated varieties and their F1´s, and the impact of the biotechnology and the mixed models on the development of cultivars. A summary of results from the researches developed in the maize breeding project at the Department of Genetics of the Agriculture College "Luiz de Queiroz" will be presented.

Linhagens elites de milho (Zea mays L.) desenvolvidas em programas genealógicos tendem a ser geneticamente relacionadas. Portanto, é necessário incorporar linhagens não relacionadas a estes programas para permitir a liberação contínua de híbridos simples superiores. Comparou-se a utilidade de um procedimento modificado de seleção recorrente recíproca (SRRM) em melhorar populações a serem utilizadas como fontes de linhagens elites e híbridos simples superiores para integrar os programas de melhoramento, e investigar os efeitos da seleção na contribuição relativa da capacidade geral (CGC) e da capacidade específica (CEC) de combinação para a variabilidade dos híbridos simples. Oito e seis linhagens S3 obtidas das populações IG-3-C1 e IG-4-C1, respectivamente, selecionadas do primeiro ciclo de SRRM, foram cruzadas no delineamento dialelo parcial e os 48 híbridos simples experimentais (HSE) e cinco híbridos simples comerciais (HSC) foram avaliados em seis ambientes. A média geral de produção de grãos dos HSE (9.57 t ha¹) não diferiu significativamente dos HSC (9.86 t ha¹), e dez dos 48 HSE poderiam ser liberados como cultivares, pois são comparáveis aos híbridos simples comerciais. Portanto, um ciclo de SRRM foi eficiente em melhorar as populações permitindo a produção de híbridos simples superiores, mas ciclos adicionais de seleção deverão ser conduzidos, pois nenhum dos HSE superou o híbrido simples comercial mais produtivo. As contribuições relativas da CGC e da CEC podem ter sido afetadas pela SRRM, uma vez que a CEC foi mais importante que a CGC para alguns caracteres avaliados.

Maize (Zea mays L.) elite inbred lines developed from pedigree programs tend to be genetically related. Therefore, it is necessary to incorporate unrelated inbreds to those programs to allow the continued release of outstanding single-crosses. The objectives of this research were to compare the usefulness of a modified reciprocal recurrent selection procedure (MRRS) to improve populations to be used as sources of elite inbreds and outstanding single-crosses to integrate pedigree programs, and to investigate the effects of selection on the relative contribution of general (GCA) and specific combining (SCA) abilities to the single-crosses variation. Eight and six S3 lines from populations IG-3-C1 and IG-4-C1, respectively, selected from the first cycle of the MRRS program were crossed in a partial-diallel mating design, and the 48 experimental and five commercial single-crosses were evaluated in six environments. Grain yield mean of the experimental single-crosses (9.57 t ha¹) did not differ from the commercial single-crosses (9.86 t ha¹), and ten of the 48 experimental single-crosses could be released as cultivars because they compared favorably to the currently used single-crosses. Thus, one cycle of the MRRS procedure improved efficiently the populations allowing the development of outstanding single-cross, but additional cycles of selection should be carried out since none of the experimental single-crosses outperformed the highest yielding commercial single-cross. The relative contribution of the GCA over SCA may have been affected by the MRRS, since the SCA was more important than GCA for some of the traits assessed.

Mapas genéticos saturados são utilizados para um eficiente mapeamento de caracteres de interesse agronômico (QTL) e nos programas de seleção assistida. Este trabalho gerou um mapa genético saturado utilizando 139 marcadores moleculares do tipo microssatélites em 256 plantas F2 geradas pelo cruzamento de duas linhagens de milho tropical (L-02-03D e L-20-01F). O mapa obtido teve uma extensão total de 1.858,61 cM, ao longo de 10 grupos de ligação, com intervalo médio entre os marcadores de 13,47 cM. Setenta e nove percento dos "bins" do mapa genético de milho foram cobertos, com um acréscimo de 14% de cobertura em relação aos mapas de milho publicados. Os resultados mostram um mapa genético mais detalhado e informativo em uma população de milho tropical representando uma primeira etapa que possibilitará desenvolver estudos da arquitetura genética para a identificação e mapeamento de QTL e a estimativa de seus efeitos sobre a variação de um caráter quantitativo, permitindo assim a sua manipulação e utilização em programas de melhoramento do milho.

Dense molecular genetic maps are used for an efficient quantitative trait loci (QTL) mapping and in the marker-assisted selection programs. A dense genetic map was generated with 139 microsatellite markers using 256 F2 plants generated by the crossing of two tropical maize inbred lines (L-02-03D and L-20-01F). This map presented 1,858.61 cM in length, where 10 linkage groups were found spanned, with an average interval of 13.47 cM between adjacent markers. Seventy seven percent of the maize genetic mapping bins were covered, which means an increase of 14% coverage in relation to the previous tropical maize maps. The results provide a more detailed and informative genetic map in a tropical maize population representing the first step to make possible the studies of genetic architecture to identify and map QTL and estimate their effects on the variation of quantitative traits, thus allowing the manipulation and use in tropical maize breeding programs.

Breeding programs for acid-soil tolerance are desirable as a relatively inexpensive and permanent way for increasing maize (Zea mays L.) yield on these soils. Our objective was to compare the genetic effects controlling the expression of maize traits in acid and non-acid soils. Seven related and one unrelated inbred lines, with different levels of tolerance to acid soil, and their F1, F2, BC1, and BC2 generations were evaluated in four acid and two non-acid soils. Estimates of additive, dominance, and epistatic effects were computed for grain yield, plant height, days to mid-silk, and prolificacy, using the generation means analysis procedure. For all traits the major part of the variation was accounted for by additive and dominance effects, with dominance effects being more important than additive and epistatic effects for both acid and non-acid soils. Epistatic effects were significant for some crosses only, being more pronounced for plant height than for the other traits. Furthermore, epistatic effects were randomly distributed among the crosses and were not related to the grain yield of the single-crosses (F1's) and to the genetic relationships of the inbreds in either type of soil. The results suggest that similar pooled gene effects control the expression of the traits assessed in both acid and non-acid soils.

A microsatellite marker (SSR) was chosen to simulate a target allele and three criteria (02, 04 and 06 markers per chromosome) were tested to evaluate the most efficient parameters for performing marker-assisted backcross (MAB) selection. We used 53 polymorphic SSRs to genotype 186 BC1 maize (Zea mays L.) plants produced by crossing the inbred maize lines L-08-05 (donor parent) and L-14-4B (recurrent parent). The second backcross (BC2) generation was produced with 180 plants and screened with markers which were not recovered from the first backcross (BC1) generation. A total of 480 plants were evaluated in the third backcross (BC3) generation from which 48 plants were selected for parental genotype recovery. Recurrent genotype recovery averages in three backcross generations were compatible with those expected in BC4 or BC5, indicating genetic gain due to the marker-assisted backcrossing. The target marker (polymorphic microsatellite PHI037) was efficiently transferred. Six markers per chromosome showed a high level of precision for parental estimates at different levels of maize genome saturation and donor alleles were not present in the selected recovered pure lines. Phenotypically, the plants chosen based on this criterion (06 markers per chromosome) were closer to the recurrent parent than any other selected by other criteria (02 or 04 markers per chromosome). This approach allowed the understanding that six microsatellites per chromosome is a more efficient parameter than 02 and 04 markers per chromosome for deriving a marker-assisted backcross (MAB) experiment in three backcross generations.

This paper reports the effects of three cycles of reciprocal recurrent selection (RRS) on the means, genetic variances, and on the genetic correlations for several traits in the IG-1 and IG-2 maize (Zea mays L.) populations. Interpopulation full-sib progenies from cycle zero (C0) and from cycle 3 (C3) of RRS were evaluated in two locations. RRS was highly effective to improve the traits according the objectives of the program: grain yield and prolificacy increased significantly, while plant height, ear height, and ear placement decreased significantly. Genetic variances for all traits decreased significantly from C0 to C3, but the genetic correlations did not change consistently across the cycles of selection. The expected responses to the fourth cycle of RRS and the probability of selecting double-crosses from C3 that outperform those from C0 showed that the decreases in the genetic variances were not great enough to limit the continued improvement of the populations as well as the use of the improved populations as sources of inbred lines to develop commercial hybrids. However, if the magnitudes of the genetic variances continue to decrease, new sources of improved germplasm should be incorporated into both populations to allow the continued improvement of the interpopulation by RRS.

The objective of this study was to evaluate whether different similarity coefficients used with dominant markers can influence the results of cluster analysis, using eighteen inbred lines of maize from two different populations, BR-105 and BR-106. These were analyzed by AFLP and RAPD markers and eight similarity coefficients were calculated: Jaccard, Sorensen-Dice, Anderberg, Ochiai, Simple-matching, Rogers and Tanimoto, Ochiai II and Russel and Rao. The similarity matrices obtained were compared by the Spearman correlation, cluster analysis with dendrograms (UPGMA, WPGMA, Single Linkage, Complete Linkage and Neighbour-Joining methods), the consensus fork index between all pairs of dendrograms, groups obtained through the Tocher optimization procedure and projection efficiency in a two-dimensional space. The results showed that for almost all methodologies and marker systems, the Jaccard, Sorensen-Dice, Anderberg and Ochiai coefficient showed close results, due to the fact that all of them exclude negative co-occurrences. Significant alterations in the results for the Simple Matching, Rogers and Tanimoto, and Ochiai II coefficients were not observed either, probably due to the fact that they all include negative co-occurrences. The Russel and Rao coefficient presented very different results from the others in almost all the cases studied and should not be used, because it excludes the negative co-occurrences in the numerator and includes them in the denominator of their expression. Due to the fact that the negative co-occurrences do not necessarily mean that the regions of the DNA are identical, the use of coefficients that do not include negative co-occurrences was suggested.

No melhoramento de milho (Zea mays L.), a utilização de cruzamentos dialélicos visando identificação de combinações superiores é uma prática comum. Esta metodologia visa estimar a capacidade de combinação dos materiais avaliados. Neste trabalho, cinco linhagens endogâmicas foram avaliadas quanto às capacidades gerais (CGC) e específica (CEC) de combinação, utilizando o esquema de dialelo completo. Os híbridos simples produzidos entre estas linhagens foram avaliados em sete ambientes, juntamente com duas testemunhas, tendo-se utilizado o delineamento de blocos casualizados. Avaliaram-se os caracteres: produção de grãos, altura da planta, altura da espiga, posição relativa da espiga e prolificidade. Realizou-se a análise dialélica, seguindo uma adaptação do método IV de Griffing, além das análises de estabilidade e adaptabilidade dos híbridos. Detectaram-se diferenças significativas para tratamentos e ambientes para todos os caracteres. A interação genótipos por ambientes foi significativa para maioria dos caracteres. As CGCs foram significativas para todos os caracteres, enquanto que a CEC foi não significativa apenas para posição relativa da espiga. Para o caráter produção de grãos, tanto os efeitos aditivos (CGC) quanto não aditivos (CEC) foram importantes, enquanto que para os demais caracteres, houve predominância dos efeitos aditivos. O híbrido mais produtivo foi obtido entre as linhagens L-08-05F e L-38-05D. Suas linhagens apresentaram elevadas CGCs e seu híbrido, elevada CEC, além de ter sido responsivo à melhoria das condições ambientais e razoavelmente estável. O segundo híbrido mais produtivo, L-46-10D x L-08-05F, mostrou ampla adaptabilidade e estabilidade.

The utilization of diallel crosses for identification of superior combinations is a common practice in maize (Zea mays L.) breeding programs. This methodology allows the estimation of the combining ability of genotypes being evaluated. In this work, five inbred lines were evaluated as to their general (GCA) and specific (SCA) combining abilities, by using a complete diallel scheme. The single-crosses produced between these inbred lines were evaluated in seven environments, along with two checks, by using a randomized complete block design. Traits analized were: grain yield, plant height, ear height, ear placement, and prolificacy. A diallel analysis was carried out, following an adaptation of Griffing's method IV, in addition to hybrid stability and adaptability analyses. Significant differences were detected for entries and environments for all traits. The interaction genotype vs. environment was significant for all traits. GCA's were significant for all traits, while SCA's were non-significant only for ear placement. For grain yield, both additive (GCA) and non-additive (SCA) effects were important, while for the remaining traits additive effects were more important. The high yielding single-cross was obtained from the cross of lines L-08-05F and L-38-05D. Those inbred lines showed higher GCA's and their cross also had high SCA; also, it is responsive to environment improvements and reasonably stable. The second in rank high yielding single-cross, L-46-10D x L-08-05F, showed wide adaptability and stability.

O milho (Zea mays L.) é uma das culturas em que a variabilidade genética tem sido extensivamente estudada com base em locos isoenzimáticos. A variabilidade genética das populações de milho BR-105 e BR-106, e dos sintéticos IG-3 e IG-4, obtidos após um ciclo de seleção recorrente de elevada intensidade, foi investigada para sete locos isoenzimáticos. Foram identificados 20 alelos, sendo que a maioria dos alelos exclusivos foi detectada na população BR-106. Um ciclo de seleção recorrente recíproca (RRS) causou reduções de 12% no número de alelos em ambas populações. Mudanças nas freqüências alélicas foram observadas entre populações e sintéticos, principalmente para o loco Est 2. As populações mostraram similaridade para número de alelos por loco, porcentagem de locos polimórficos, e para heterozigosidade observada e esperada. Houve decréscimo nas estimativas da variabilidade dos sintéticos em conseqüência dos efeitos da deriva genética e redução do tamanho efetivo populacional. A distribuição da diversidade genética dentro e entre as populações revelou que a maior parte da diversidade permaneceu dentro delas, i.e. BR-105 x BR-106 (G ST = 3,5%) e IG-3 x IG-4 (G ST = 4,0%). A distância genética entre as populações e os sintéticos aumentou em torno de 21%. Houve aumento na divergência genética entre as populações, porém sem comprometer novos procedimentos de seleção.

Maize (Zea mays L.) is one of the crops in which the genetic variability has been extensively studied at isoenzymatic loci. The genetic variability of the maize populations BR-105 and BR-106, and the synthetics IG-3 and IG-4, obtained after one cycle of a high-intensity reciprocal recurrent selection (RRS), was investigated at seven isoenzymatic loci. A total of twenty alleles were identified, and most of the private alleles were found in the BR-106 population. One cycle of reciprocal recurrent selection (RRS) caused reductions of 12% in the number of alleles in both populations. Changes in allele frequencies were also observed between populations and synthetics, mainly for the Est 2 locus. Populations presented similar values for the number of alleles per locus, percentage of polymorphic loci, and observed and expected heterozygosities. A decrease of the genetic variation values was observed for the synthetics as a consequence of genetic drift effects and reduction of the effective population sizes. The distribution of the genetic diversity within and between populations revealed that most of the diversity was maintained within them, i.e. BR-105 x BR-106 (G ST = 3.5%) and IG-3 x IG-4 (G ST = 4.0%). The genetic distances between populations and synthetics increased approximately 21%. An increase in the genetic divergence between the populations occurred without limiting new selection procedures.

A proteína do milho (Zea mays L.) é considerada como de baixa qualidade, pois apresenta nível reduzido dos aminoácidos essenciais lisina e triptofano. Uma alternativa para contornar esse problema consiste na introdução do gene opaco-2, que eleva a quantidade desses aminoácidos, embora apresente efeitos pleiotrópicos negativos em caracteres agronômicos relacionados à produtividade. O objetivo deste trabalho foi verificar a viabilidade de melhorar a qualidade protéica do milho com uso de Seleção Recorrente Fenotípica (SRF), sem introdução do gene opaco. Foram realizados quatro ciclos de SRF em duas populações, IG-1 e IG-2, para o teor de triptofano nos grãos, com 20% de intensidade. Os quatro ciclos seletivos e as populações originais foram avaliados para caracteres agronômicos em três locais. Para avaliação da qualidade protéica, foi conduzido um experimento com controle da polinização, para se evitar o efeito de xênia. Houve pequeno aumento nos níveis de triptofano para IG-1 (cerca de 0,70% por ciclo) e ausência de alteração em IG-2. Ocorreu aumento na relação triptofano/proteína para IG-1 (1,26% por ciclo) e os teores de proteína não se alteraram para as duas populações. A qualidade protéica da testemunha ESALQ VD2-opaco foi superior a das populações mesmo após a realização dos quatro ciclos. Como resposta correlacionada à seleção, houve redução na produção de grãos (2,50% por ciclo), prolificidade, altura da planta e altura da espiga. O baixo ganho, associado às alterações desfavoráveis em caracteres agronômicos, indica que este método de seleção possivelmente não é eficiente para elevar a qualidade protéica.

Maize (Zea mays L.) protein is considered to be of low quality due to low levels of the essential lysine and tryptophan amino acids. An alternative to solve this problem is to use the opaque-2 gene, which improves the level of these amino acids, but has negative pleiotropic effects on agronomic characters. A phenotypic recurrent selection scheme was carried out in two non-opaque maize populations to verify the possibility of improving their protein quality without using this gene. Four cycles were completed and a 20% selection intensity for tryptophan content in the kernels was used in two populations, IG-1 and IG-2. The original and the four-cycle populations were evaluated in three locations for agronomic traits. For protein and tryptophan content, a separated trial was carried out because plants of the plots were hand-pollinated. No increase in tryptophan content was observed in the IG-2 population, whereas IG-1 presented a small increase (0.70% per cycle). The ratio tryptophan/protein increased 1.26% per cycle in IG-1 and the protein content did not increase in both populations. The ESALQ-VD2-opaque check was superior in relation to both populations for protein quality, as expected, even after completion of four selection cycles. The kernel yield (2.5% per cycle) prolificacy, plant and ear heights, decreased with selection cycles, as a correlated response to selection. Phenotypic recurrent selection in non-opaque maize was not able to increase, at reasonable rates, the protein quality of maize kernels.

Os grupos heteróticos são importantes no melhoramento de milho, pois permitem o uso mais eficiente do germoplasma. Assim, o objetivo deste trabalho foi alocar linhagens de milho em grupos heteróticos a partir de estimativas da capacidade específica de combinação. Oito linhagens S3 da população BR-105 e dez da BR-106, foram cruzadas ao nível interpopulacional seguindo um sistema dialélico. Oitenta híbridos simples foram obtidos e avaliados em látices em três ambientes. Foram avaliados os caracteres produção de grãos (PG), altura da planta (AP) e altura da espiga (AE). As estimativas das capacidades geral (CGC) e específica (CEC) de combinação foram obtidas segundo o método 4, modelo I de Griffing. A partir das estimativas de CEC, as linhagens foram alocadas aos respectivos grupos heteróticos utilizando-se o método UPGMA (média aritmética não ponderada) para a construção do dendrograma e a dispersão gráfica pelo método das coordenadas principais. Para PG, as linhagens foram alocadas em quatro grupos heteróticos, sendo dois grupos em cada população. Para AP e AE, o uso das estimativas de CEC não foi eficiente para alocar as linhagens em grupos heteróticos. As análises de agrupamento e de coordenadas principais foram eficientes na alocação das linhagens em grupos heteróticos para PG. Então, como a produção de grãos é a principal característica para o melhoramento de milho, com a alocação das linhagens em quatro grupos heteróticos, os cruzamentos serão direcionados e o processo de melhoramento se tornará mais eficiente, evitando a obtenção e a avaliação de cruzamentos (híbridos) desnecessários.

The heterotic groups are important in maize breeding programs because they allow the most efficient use of the germoplasm. The objective of this research was to allocate maize lines to heterotic groups from estimates of specific combining ability (SCA). Eight and ten S3 lines derived from populations BR-105 and BR-106, respectively, were crossed at interpopulation levels following a diallel system. Eighty single-crosses were obtained and evaluated in lattice designs across three environments. Recorded data consisted of grain yield (GY), plant height (PH) and ear height (EH). Estimates of general and specific combining ability (GCA) and (SGA) were computed according to Griffing's method 4, model I, for all traits. Estimates of SCA were used for principal coordinate analysis (PCO) and for cluster analysis, by using UPGMA (unweighted pair-group method with arithmetical averages) clustering algorithm, to assign the lines to heterotic groups. For the trait GY the lines were allocated to 4 heterotic groups, in which the lines from each population were subdivided in 2 heterotic groups. For the traits PH and EH, the use of the estimates of SCA to assign lines to heterotic groups were not efficient. Cluster analysis and principal coordinate analysis were efficient for the trait GY in allocating maize lines to heterotic groups. Thus, since grain yield is the main trait for the breeding purposes, with the allocation of the lines in 4 heterotic groups, the crosses will be more efficient, avoiding to obtain and evaluate unnecessary (hybrid) crosses.

Foi derivada uma expressão para predizer a resposta à seleção recorrente para tamanhos efetivos populacionais (Ne) pequenos. Usualmente, as respostas à seleção têm sido estimadas utilizando-se da expressão Rs = ics²A/sPh, onde i, c, s²A e sPh referem-se ao diferencial de seleção estandardizado, controle parental, variância genética aditiva e desvio-padrão fenotípico, respectivamente. Entretanto, essa expressão foi derivada assumindo que o tamanho efetivo populacional é infinito. Considerando que a população sob seleção é finita, derivou-se uma nova expressão considerando o tamanho efetivo populacional finito: Rs = [ic(sigma²A + deltaFD1)/ sigmaPh) - deltaFID], onde deltaF, ID, e D1 referem-se às alterações no coeficiente de endogamia, à depressão por endogamia, e à covariância dos efeitos aditivos e de dominância dos homozigotos, respectivamente. Portanto, as respostas esperadas à seleção baseadas nesta expressão serão inferiores àquelas obtidas na expressão padrão, principalmente para caracteres com elevados níveis de dominância como produção de grãos. Foram estimadas as respostas à seleção recorrente com progênies de meios-irmãos para uma população de milho utilizando-se as duas expressões, considerando-se a avaliação de 100 progênies e recombinação de 10 progênies S1's, o que corresponde a um tamanho efetivo de Ne = 10 para cada ciclo. A resposta à seleção acumulada estimada com a nova expressão foi cerca de 47 e 28% inferior para produção de grãos e altura da planta, respectivamente, que aquela obtida com a expressão usual que não considera o tamanho efetivo populacional. Portanto, a expressão padrão que tem sido utilizada superestima as respostas à seleção recorrente, como tem sido reportado na literatura, por não considerar o tamanho efetivo populacional normalmente baixo nestes tipos de programas.

A formula was derived for the prediction of the response to recurrent selection when the effective population size (Ne) is small. Usually, responses to selection have been estimated by Rs = icsigma²A/sigmaPh, where i, c, sigma²A, and sigmaPh stand for standardized selection differential, parental control, additive variance, and phenotypic standard deviation, respectively. This expression, however, was derived under the assumption of infinite population size. By introducing the effects of finite population size, the expression derived was Rs = [ic(sigma²A + deltaFD1)/sigmaPh] - DFID, where deltaF, ID and D1 are the changes in the inbreeding coefficient, the inbreeding depression, and the covariance of additive and homozygous dominance effects, respectively. Thus, the predicted responses to selection based on these expressions will be smaller than those based on the standard procedures for traits with a high level of dominance such as yield. Responses to five cycles of half-sib selection were predicted for maize by both expressions, considering that 100 progenies were evaluated and 10 S1 progenies were recombined, which corresponds to Ne = 10 for each cycle. The accumulated response to selection estimated with the new expression was about 47 and 28% smaller than that based on the standard expression for yield and plant height, respectively. Thus, the expression usually used overestimates the responses to selection, which is in agreement with reported results, because it does not take into account the effective population size that is generally small in recurrent selection programs
